My research work focused on understanding the dysregulation in neuronal circuitry leading to neurodegenerative disease, using zebrafish as a model organism. During my master’s project at IIT Hyderabad and working on eye-retina diseases at LV Prasad Eye Institute, I was exposed to the idea of working on neurodegenerative diseases. My master's thesis was focused to determine the quantitative heterogeneity by G-protein coupled receptors (GPCRs) mediated Ca2+ dynamics in live cells using time-lapse confocal microscopy which also involved computational biology and statistical data analysis by MATLAB and writing algorithms. I continued my research on neurodegenerative diseases in my doctoral studies at the Medical University of Warsaw, Poland. I have investigated the role of the stim2a in the model of neurodegenerative disorders using scRNA-seq, Ca2+ imaging in live animals, performing behavioral experiments to identify the change in cognitive ability, molecular biology techniques, and biological data analysis and simulation using MATLAB and R-programming. I discovered that the knockout of the stim2a in zebrafish affects the Ca2+ homeostasis in brain neurons. I also found significant changes in the differential expression of some important genes, which mediate Ca2+ signaling, affecting neuronal circuitry. In addition, behavioral studies showed that knockout of stim2a induced hyperactive-like behavior in zebrafish.
